與二抗（或生物素化一抗的鏈霉親和素-HRP）孵育

13. 加入 100 μl 標(biāo)記的二抗。要添加的量可以在初步實(shí)驗(yàn)中確定。為了準(zhǔn)確定量，應(yīng)過(guò)量使用標(biāo)記的二抗。所有稀釋都應(yīng)在稀釋緩沖液中進(jìn)行。

14. 用粘性塑料蓋住板，在室溫或 -20°C 下孵育 60-90 分鐘。

17. 在建議的孵育時(shí)間過(guò)后，向每個(gè)孔中加入終止溶液。

19. 計(jì)算每組重復(fù)標(biāo)準(zhǔn)品、樣品和對(duì)照品的平均吸光度值。

20. 如果單個(gè)吸光度值與相應(yīng)的平均值相差超過(guò)15%，則認(rèn)為結(jié)果可疑，應(yīng)重新測(cè)定樣品。

21. 使用能夠生成良好曲線擬合的計(jì)算機(jī)軟件減少數(shù)據(jù)，從而創(chuàng)建標(biāo)準(zhǔn)曲線。

22. 如果樣品已被稀釋，則根據(jù)標(biāo)準(zhǔn)曲線測(cè)定的濃度必須乘以稀釋因子。
